Cancer is a complex disease characterized by dysregulated cellular processes and resistance to apoptosis. Caspase-8 and caspase-10, key players in the apoptotic signaling pathway, play crucial roles in regulating cell death and preventing tumor growth. However, in many cancer types, these caspases are often downregulated or inactivated, leading to impaired apoptosis and enhanced tumor survival. This chapter aims to explore the potential of reinstating the expression and activation of caspase-8 and caspase-10 as a therapeutic strategy for cancer treatment. We discuss the molecular mechanisms underlying the dysregulation of caspase-8 and caspase-10 in cancer and highlight recent advancements in targeting these caspases to induce tumor cell death. Furthermore, we examine the challenges and opportunities associated with targeting caspase-8 and caspase-10, including potential side effects and the development of resistance. Overall, this chapter emphasizes the importance of caspase-8 and caspase-10 in cancer therapy and provides insights into novel therapeutic approaches that exploit the restoration of their expression and activation.
